Forskolin is a diterpene from Coleus forskohlii that directly activates adenylyl cyclase, increasing intracellular cAMP — a fundamental signaling molecule. Our research shows one notable study where 250mg of 10% forskolin extract twice daily increased testosterone by 16.77% and improved body composition in overweight men. However, the evidence base is thin (primarily one study), and forskolin's potent cAMP activation means it can significantly affect blood pressure, heart rate, and drug metabolism.

WHAT IT DOES

Forskolin bypasses all cell surface receptors and directly activates adenylyl cyclase, the enzyme that converts ATP to cyclic AMP (cAMP). cAMP is a universal "second messenger" that activates protein kinase A, which then: stimulates lipolysis (fat burning), increases thyroid hormone production, enhances testosterone synthesis in Leydig cells, relaxes smooth muscle (bronchodilation, vasodilation), and reduces intraocular pressure. The problem is this mechanism is extremely broad — increasing cAMP affects virtually every cell type, which is why the interaction profile is extensive.

OPTIMAL DOSAGE

  • Look for: Coleus forskohlii extract standardized to 10% forskolin, 250mg per serving, ForsLean branded extract
  • Avoid: Unstandardized coleus root powder; products with <10% forskolin; "fat burner" blends with undisclosed forskolin amount
  • Minimum effective dose: 25mg forskolin 2x/day (from 250mg of 10% extract)
